小麦抗吸浆虫SSR标记开发和应用

摘要:
麦红吸浆虫(Sitodiplosis mosellana Géhin)是严重威胁小麦生产的重要害虫,开发与抗虫基因紧密连锁或共分离的标记将为利用抗虫基因资源改良小麦抗虫性提供方法。本研究在对抗麦红吸浆虫主效QTL Qsm.hbau-4A定位基础上,试图从QTL所在区间内开发SSR标记,并利用这些标记对一套小麦种质资源进行了标记基因型鉴定。结果表明,所开发的565个SSR标记中,12%的标记在小麦亲本间、2%的标记在3套近等基因系(NILs)间有多态性。在对不同抗性的重组近交系(RILs)进行标记基因型检测时,这些SSR标记平均符合率是90.35%;在对不同抗性种质资源的平均检测有效率是51.04%,其中以Xhs87201bp标记的检测有效率最高(65.37%),可以用于抗虫种质资源筛选筛选。与本课题组前期定位或开发的标记相比,新开发标记的检测有效率高,且多为共显性标记,可以用于对小麦抗虫性种质资源的分子标记选择和抗虫主效QTL的精细定位。

Development and Application of SSR Markers Surrounding the OWBM-Tolerant Locus Qsm.hbau-4A in Wheat

Abstract:
Orange wheat blossom midge (OWBM, Sitodiplosis mosellana Gehin) is an important pest insect that seriously threatens wheat production. Development of markers that closely linked to or co-segregated with OWBM resistance is of importance in genetic dissection of the major quantitative trait locus (QTL) tolerant to OWBM and marker-assisted breeding for resistant wheat varieties. Based on previous mapping of the major QTL Qsm.hbau-4A, here we attempted to develop SSR markers tightly linked with this QTL, followed by marker-based genotyping in a panel of wheat accessions. Out of the 565 SSR markers developed, 12% were detected to be polymorphic between both parents, whereas 2% were polymorphic in three pairs of near isogenic lines (NILs). Genotyping using these SSR markers revealed the average coincidence rate of 90.35% in recombinant inbred lines (RILs) with variable OWBM resistance, as well as a mean value on detection efficiency of 51.04% in 566 wheat accessions showing different OWBM resistance. The marker Xhs87201bp represented the highest detection efficiency rate (65.37%) and this might be useful in screening resistant germplasm resources. The SSR markers if relative to those produced in the former study showed mostly co-dominant and high detection efficiency. Collectively, these newly-developed markers provided a source valuable in fine mapping of Qsm.hbau-4A and marker-assisted selection of wheat OWBM resistance.
