本研究以低含量γ-氨基丁酸的宁农黑粳为母本,高含量γ-氨基丁酸的高粱稻-1 为父本,构建 F2群体,获得了 216个 F2 单株。利用 130 个 SSR 标记构建了一张 F2 群体的 SSR 标记连锁图谱,覆盖基因组长度为 2406.9cM,连锁群长度在129.5-360.7 cM,标记间的平均距离为 18.5 cM,并进一步开展控制水稻γ-氨基丁酸含量的 QTL 定位研究。结果表明:共检测到 7 个 QTL 位点,分别位于第 8 号和第 9 号染色体上,其中 qGABA8-2、qGABA8-3、qGABA9-1 的贡献率依次为 10%、11%和9%。对 3 个贡献率大的 QTL 位点进行复合区间作图,当 LRS 为 25.6 时,在 RM342-RM515 处可能存在较为可靠的 QTL,初步将qGABA8 定位在标记 RM342 与 RM515 之间的 326Kb 区间内。利用 InDel 标记对目标区间加密,将该区间进一步缩小到 183 Kb区间内,位于标记 RM342 和 G121 之间。本研究结果可进一步通过构建次级群体对该基因进行精细定位及图为克隆,同时,研究中筛选出的 SSR 标记和设计的 InDel 标记可快速筛选水稻育种材料中富γ-氨基丁酸的基因型,加快育种进程。
The rice cultivars Ningnong black carp and high indica rice exhibited low and high content on γ-aminobutyric acid, respectively. Both genotypes were deployed for generating a F2 population that comprises of 216 individual plants. By taking use of 130 polymorphic SSR markers, a linkage map was produced with an length of 2406.9 cM. Each linkage group expands from 129.5 to 360.7 cM, and a mean distance between markers was 18.5 cM. The QTL mapping positioning study showed that seven QTL loci controlling the γ-aminobutyric acid content were found on chromosomes 8 and 9, respectively. The contribution rate of qGABA8-2, qGABA8-3, qGABA9-1 was 10%, 11% and 9%, respectively. The composite interval mapping was performed on three QTL sites with large contribution rate. When LRS was 25.6, a QTL qGABA8 was initially found at RM342-RM515 in an interval of 326Kb. The confidence interval was further delimited to 180 kb by using the InDel marker G121. Therefore, the accumulated results of this study might be useful in further gene isolation and marker-assisted screening for the genotypes with the enriched amino butyric acid.
