朝鲜淫羊藿(Epimedium koreanum Nakai)作为中国东北地区重要的药用植物,因过度采挖导致野生资源锐减,因此研究其种质资源的遗传多样性,并解析其居群间的亲缘关系对其保护策略的制定具有重要意义。采用ISSR(Inter simple sequence repeat)标记技术,前期通过优化反应体系筛选出7条高多态性引物,对采集自东北地区14个居群的128份样本进行了遗传多样性分析。结果表明,7条ISSR引物共扩增出80个条带,其中74个条带表现出多态性,多态性条带比例达到92.60%;平均等位基因数为1.6438,平均有效等位基因数为1.3747,平均Nei′s基因多样性指数为0.2189,平均Shannon′s信息指数为0.3290,其中WNS居群(辽宁省桓仁县五女山景区)遗传多样性最高,而DLZC居群(吉林省临江市大栗子村)最低;与同属植物进行比较后发现朝鲜淫羊藿种质资源具有中等水平的遗传多样性。居群间的遗传相似系数范围在0.7441~0.9700之间,遗传距离在0.0304~0.2955;Mantel检验显示遗传距离与地理距离无显著相关性,表明居群分化可能受生境隔离或繁殖特性影响。遗传结构分析(Gst=0.2784)表明,72.16%的遗传变异发生在群体内。此外,通过聚类分析将14个居群划分为5个类群。本研究为朝鲜淫羊藿的遗传多样性保护及种质资源管理提供了数据基础。
Epimedium koreanum Nakai, a medicinal plant of critical importance in Northeast China, has experienced severe depletion of its wild populations due to overharvesting. Investigating the genetic diversity of its germplasm and elucidating phylogeographic relationships among populations are therefore essential for developing effective conservation strategies. The research employed ISSR (Inter simple sequence repeat) marker technology. Through preliminary optimization of the reaction system, seven highly polymorphic primers were selected for genetic diversity analysis of 128 samples collected from 14 natural populations in Northeast China. Results showed that these seven ISSR primers amplified a total of 80 bands,with 74 (92.60%) being polymorphic. Number of observed alleles (Na) is 1.6438, effective number of alleles (Ne) is 1.3747, Nei′s gene diversity index (H) is 0.2189, and Shannon′s information index (I) is 0.3290. The WNS population (Wunu Mountain Scenic Area, Huanren county, Liaoning province) showed the highest genetic diversity, while the DLZC population (Dalizi village, Linjiang city, Jilin province) exhibited the lowest. After comparison with other species in the same genus, it was found that the genetic diversity of Epimedium koreanum germplasm resources is at a moderate level. Genetic similarity coefficients among populations ranged from 0.7441 to 0.9700, with genetic distances between 0.0304 and 0.2955. Mantel test showed no significant correlation between genetic and geographical distances, suggesting that population differentiation may be influenced by habitat isolation or reproductive characteristics rather than geographical distance. Genetic structure analysis (Gst=0.2784) indicated that 72.16% of genetic variation occurred within populations. Furthermore, cluster analysis divided the 14 populations into five distinct groups. This study provides fundamental data for the conservation of genetic diversity and germplasm resource management of E. koreanum.
