转录调控辅助因子VQ蛋白在植物响应非生物胁迫方面发挥重要作用。本研究在全基因组水平对紫花苜蓿VQ基因家族进行了鉴定和系统分析。结果表明,紫花苜蓿基因组共有24个VQ基因家族成员,不均匀地分布在8条染色体上,除了MsVQ8和MsVQ17,其余MsVQ基因均无内含子结构。亚细胞定位预测显示大多数MsVQ蛋白定位于细胞核。系统发育进化关系表明,24个MsVQ家族归属于7个亚家族,且同一亚家族成员的理化性质和保守基序相似。紫花苜基因组中包含 6对MsVQ大片段重复基因,1对MsVQ串联重复基因。胁迫响应元件、激素响应元件及WRKY结合元件广泛分布在MsVQ基因的启动子区域。转录组数据分析结果表明MsVQ基因对干旱、盐及低温等非生物胁迫均有响应。qRT-PCR分析结果表明,在盐胁迫下,3个MsVQ基因表达上调。在碱处理下,6个MsVQ基因均在处理6h时表达量最高。本研究结果为进一步研究紫花苜蓿VQ基因家族生物学功能及分子育种奠定了理论基础。
VQ motif-containing proteins play important roles in plant abiotic and biotic stresses.In this study, VQ gene family were identified and systematically analyzed from Medicago sativa. A total of 24 MsVQ genes were identified in the alfalfa genome, which were unevenly distributed on eight chromosomes. Expect from MsVQ8 and MsVQ17, MsVQ genes do not contain introns and a series of MsVQs were predicted to localized at nucleus. Phylogenetic analysis showed that MsVQ was divided into seven subgroups, each subfamily shared relatively conserved gene structures and protein motifs. Medicago sativa genome included 6 pairs of segmental duplicated genes and 1 pair of tandem repeat genes. The abiotic stress-responsive elements, hormone-responsive elements and WRKY binding box were presented in the promoters of MsVQs. Transcriptome data showed that MsVQs were respond to abiotic stresses such as drought, salt and low temperature. The quantitative real-time PCR (qRT-PCR) found that three MsVQs genes were significantly induced by salt stress and six MsVQs genes expression levels reached a peak at 6h after alkaline stress. Taken together, these findings will provide clues for further study of MsVQs functions and alfalfa molecular breeding.
