为探明四川省旺苍县茶树种质资源的遗传多样性,挖掘潜在的优异种质用于新品种选育,本研究利用自研的茶树液相功能芯片对124份旺苍代表性茶树资源的76252个SNP位点进行了基因型检测,结合理化鉴定和感官审评数据,全面分析了旺苍地方资源的遗传结构和品质特征;并通过全基因组关联分析进一步挖掘了品质性状显著关联的SNP位点和候选基因。结果表明:旺苍县域内茶树资源多样性丰富且具有一定的地域性特点,其中大两古茶树资源为过渡型茶树群落,富含酯型儿茶素,尤其是EGCG,所制烘青绿茶品质优良,香气普遍高爽/鲜、有花(果)香,滋味以甘醇/和、鲜爽为主,具有极高的育种价值。根据芯片检测的40,326个核心SNP位点,发掘出4个与茶树氨基酸、茶多酚和EGCG含量高度相关的候选基因,分别是γ-CA1、TPR5、MYB94和UGT85A1。相关研究结果可为今后旺苍茶树种质资源的开发利用提供理论依据。
To clarify the genetic diversity of tea plant germplasm resources in Wangcang County, Sichuan Province, and identify potential superior germplasms for new cultivar breeding, this study utilized a self-developed liquid-phase functional microarray for tea plants to genotype 76,252 SNP loci in 124 representative tea resources from Wangcang. Combined with biochemical component detection and organoleptic evaluation, the genetic structure and quality characteristics of these local resources were comprehensively analyzed. Genome-wide association studies were further conducted to identify SNP loci and candidate genes significantly associated with quality traits. The results show that the tea germplasm resources within the county exhibit rich diversity and distinct regional characteristics. Among them, the ancient tea plant population in Daliang is identified as a transitional type, rich in esterified catechins—particularly EGCG. The baked green tea produced from these plants demonstrates excellent quality, with a generally refreshing and high-aromatic profile featuring floral (or fruity) notes, and a taste characterized by sweetness, mellowness, and freshness, highlighting its significant breeding value. Based on 40,326 core SNP loci detected, four candidate genes highly associated with the content of amino acids, tea polyphenols, and EGCG were identified: γ-CA1, TPR5, MYB94, and UGT85A1. These results provide a theoretical foundation for the future development and utilization of tea germplasm resources in Wangcang.
