    哈密大枣(Ziziphus jujuba ‘Hamidazao’)是新疆特有的地方优良品种,对其开展遗传多样性研究对哈密大枣优质种源的保护和良种选育具有重要意义。以74份哈密大枣优株为材料,对30个农艺性状进行测量,采用变异分析、聚类分析等方法进行综合评价,并用11条多态性高的ISSR引物进行PCR扩增分析。结果显示,哈密大枣优株之间的农艺性状变异系数在5.99%~30.09%之间,遗传多样性指数在1.36~1.95之间;基于表型性状聚类,在欧氏距离27处可将74份材料分为五大类群,第Ⅰ类表现出较强的营养生长势能,第Ⅱ类针刺长度显著高于其他类别,第Ⅲ类存在明显的生长弱势,第Ⅳ类呈现出均衡且偏优的生长势,第Ⅴ类为具有高产潜力的优良种质资源。11条ISSR引物在74份哈密大枣优株中共检测到94条条带,其中多态性条带75条,平均每个引物多态性条带比例为80.30%,平均观测等位基因数为1.80,平均有效等位基因数为1.35,平均Nei′s基因多样性指数为0.21,平均Shannon′s多样性信息指数为0.33。基于遗传相似系数可将哈密大枣优株分为五大类群,第Ⅰ类仅包括HM56单株,第Ⅱ类仅包括HM57,第Ⅲ类包含HM21、HM27、HM49和HM64,第Ⅳ类包括HM22和HM32,其余66份材料均归属于第Ⅴ类。优株之间遗传距离在0.04~0.52之间,遗传相似性在0.60~0.96之间,遗传相似性最高且遗传距离最小的组合为HM7与HM8、HM7与HM11;而遗传相似性最低且遗传距离最大的组合为HM3与HM56。研究表明哈密大枣优株之间具有一定的遗传变异,遗传信息相对丰富,可为未来哈密大枣资源利用和良种选育提供理论依据。

    Abstract:

    Hamidazao jujube (Ziziphus jujuba ‘Hamidazao’) is a superior local cultivar exclusively in Xinjiang, China. Investigating its genetic diversity is sigficant for conserving high-quality germplasm and breeding improved cultivars. In this study, 74 elite individuals of Hamidazao jujube were evaluated through measuring 30 agronomic traits. Comprehensive assessments were conducted via variation analysis and cluster analysis, along with PCR amplification using 11 highly polymorphic ISSR primers. The results indicated that the coefficients of variation for agronomic traits among the elite individuals ranged from 5.99% to 30.09%, while genetic diversity indices varied from 1.36 to 1.95. Cluster analysis based on phenotypic traits classified the 74 accessions into five major groups at a Euclidean distance of 27. Group I exhibited strong vegetative growth vigor; Group II had significantly longer spines than the other groups; Group III showed obvious growth weakness; Group IV exhibited balanced and relatively superior growth performance; Group V comprised germplasm with high yield potential. Through the 11 ISSR primers, 94 bands were detected across the 74 elite individuals, of which 75 were polymorphic. The mean percentage of polymorphic bands per primer was 80.30%, with an average observed allele number of 1.80, an average effective allele number of 1.35, an average Nei′s gene diversity index of 0.21, and an average Shannon′s information index of 0.33. Based on the genetic similarity coefficient, the individuals were divided into five groups: Group I contained only HM56; Group II contained only HM57; Group III included HM21, HM27, HM49, and HM64; Group IV contained HM22 and HM32; and the remaining 66 accessions fell into Group V. Genetic distances among the elite individuals ranged from 0.04 to 0.52, with corresponding genetic similarities from 0.60 to 0.96. The combinations with the highest genetic similarity and the smallest genetic distance were HM7 and HM11,HM8 and HM11, while the combination with the lowest genetic similarity and the largest genetic distance is HM3 and HM56.These results suggest that considerable genetic variation and relatively rich genetic information were found among the elite individuals of ‘Hamidazao’ jujube, providing a theoretical foundation for future germplasm utilization and breeding efforts.
