自噬途径广泛参与调控植物的生长发育、营养物质的循环利用、多种生物与非生物逆境的耐受性,但其在水稻中功能研究还处在初始阶段。本研究克隆了水稻自噬相关基因OsATG12,构建了OsATG12基因的超表达载体、基因编辑载体,进行水稻遗传转化后获得了相应的转基因材料。进一步通过分子鉴定获得了OsATG12超表达转基因株系和atg12s突变体株系;通过对这些转基因株系材料进行稻瘟病抗性鉴定发现,OsATG12基因负调控水稻对稻瘟病的抗性。这为揭示水稻自噬途径及自噬基因的功能提供了新思路。
Autophagy pathway is known to be widely involved in the regulation of growth and development, nutrient recycle, a variety of biotic or abiotic stresses tolerance in plant. However, the functional research in rice is still in its infancy. In this study, OsATG12, a rice autophagy-related gene, was isolated and subjected for producing over-expressing and gene editing plasmids transformed into rice. We identified the transgenic lines over-expressing OsATG12 and atg12s mutant lines. By testing for rice blast resistance using these transgenic lines, OsATG12 was found to negatively regulate the blast resistance of rice. Collectively, this study provided new insights for revealing the functions of rice autophagy pathway and autophagy related genes.
